一般描述
邻苯二胺(o-PD)是一种无色的非荧光分子。
邻苯二胺二盐酸盐(OPD)是一种显色底物,因为利用辣根过氧化物酶缀合物,因此可用于酶联免疫吸附试验(ELISA)程序。底物产生可溶的最终产物,其颜色为橙棕色,并且可以在450 m下通过分光光度计读取。可以用3N HCl或3M H2SO4终止OPD反应,并在492 nm处读取。
应用
在酶联免疫吸附试验(ELISA)中,邻苯二胺盐酸盐用作辣根过氧化物酶偶联二抗的底物。
重悬
将一片药片溶于 pH 5.0 的 0.05M 磷酸盐-柠檬酸盐缓冲液中至所需浓度(通常使用0.4mg/ml 的 OPD 浓度)。使用前,在每 100ml 底物缓冲溶液中加入 40 μl 新鲜的 30% 过氧化氢,现配现用。片剂分别包装在箔包内。
