碱性磷酸酶可与抗体及其他蛋白质偶联而用于ELISA、Western blotting和组织化学检测。它通常用于使蛋白质和核酸去磷酸化。当需要高灵敏度时,可用于蛋白标记。碱性磷酸酶也可用于使DNA或RNA的5′-末端去磷酸化以防止自连。在用碱性磷酸酶去磷酸化后,DNA或RNA也可用放射性标记的磷酸盐(通过T4多核苷酸激酶)进行标记。
生化/生理作用
碱性磷酸酶可使蛋白质和核酸去磷酸化。人胎盘碱性磷酸酶(PLAP)具有独特的残基Glu-429。它是一种可被L-Phe、L-Trp和L-Leu无竞争性抑制的变构酶。PLAP是经过翻译后修饰的。磷脂酰肌醇特异性磷脂酶C可能参与到了PLAP的释放。PLAP可能在细胞分裂中发挥作用,并参与搭配母体IgG向胎儿的转移。
