Sigma′s Nitric Oxide Synthase Detection System is a simple and specific assay for the measurement of nitric oxide synthase (NOS) activity. Nitric oxide (NO), formed from the amino acid L-arginine via the action of the NOS enzyme, is a unique cell signaling molecule that functions as both an intracellular and an extracellular messenger. NO has been implicated in vasodilation, neurotransmission, cytotoxicity, and inflammation.1
This kit utilizes a highly specific and sensitive assay for the direct quantitative measurement of nitric oxide synthase activity. The assay measures the enzymatic conversion of 3H-arginine to 3H-citrulline and is the standard assay for measuring NOS activity in cell lysates, tissue homogenates and purified enzyme preparations. Activity is measured in the picomole range and a positive control is included.

Nitric oxide, which mediates influences of numerous neurotransmitters and modulators on vascular smooth muscle and leukocytes, can be formed in the brain from arginine by an enzymatic activity that stoichiometrically generates citrulline. We show that glutamate and related amino acids

Research on the biological roles of nitric oxide has revealed that it functions as an important signal and effector molecule in a variety of physiologic and pathologic settings. In animals, nitric oxide is synthesized enzymatically from L-arginine through the actions
Cancer metastasis reviews, 17(1), 7-23 (1998-04-17)
Nitric oxide (NO) is a potent biologic mediator with diverse physiologic and pathophysiologic roles. NO is produced from L-arginine by the family of nitric oxide synthase (NOS) enzymes, forming the free radical NO and citrulline as byproduct. Three distinct isoforms
Current opinion in chemical biology, 2(5), 656-663 (1998-11-18)
The enzyme nitric oxide synthase catalyzes the oxidation of the amino acid L-arginine to L-citrulline and nitric oxide in an NADPH-dependent reaction. Nitric oxide plays a critical role in signal transduction pathways in the cardiovascular and nervous systems and is
