应用

制霉菌素是一种多烯类抗生素。 用途包括预防培养细胞污染,诱导表达TLR2的THP1细胞分泌白介素(IL)-1、IL-8和肿瘤坏死因子α。 该产品的推荐细胞培养使用浓度为50 mg/L。

生化/生理作用

作用机制:制霉菌素通过与真菌细胞膜中的麦角甾醇结合,增加膜的渗透性和产生基于制霉菌素/麦角甾醇的离子通道起作用。 它还可作为脂筏抑制剂和膜相关胆固醇。

抗菌谱:制霉菌素可对抗真菌、酵母和霉菌。

注意

制霉菌素的组织培养基溶液在37°C下可稳定保存三天。 在中等碱性介质中也很稳定。 溶液和水悬液在制备后不久就开始失去活性,热、光和氧气会加速分解。 如果在pH 7下加热至100°C,水悬液可稳定保存10 min。

制备说明

在28 °C,制霉菌素可溶于甲醇(11.2 mg/mL)、乙醇(1.2 mg/mL)、四氯化碳(1.23 mg/mL)、氯仿(0.48 mg/mL)、苯(0.28 mg/mL)和乙二醇(8.75 mg/mL)。 也可以5 mg/mL溶于DMSO,易溶于DMF和甲酰胺。 不建议对制霉菌素溶液进行高压灭菌或无菌过滤。 制霉菌素混悬液也有效,37 °C下可稳定保存3天。 制备50 mg/mL的储备混悬液,-20°C下保存。对N6261进行γ-辐照得该产品。

将容器密闭保存在干燥和通风良好的地方。在惰性气体下保存。对空气和光敏感。在干燥处保存。
