过氧化物(如过氧化氢H2O2)是分子氧代谢产生的活性氧类,可作为细胞内信使和氧化应激来源。细胞可通过多种机制产生过氧化氢,如嗜中性粒细胞和巨噬细胞(呼吸爆发)中由NOX介导的ROS形成过程,或在线粒体呼吸的电子泄漏过程中由超氧阴离子歧化酶生成。 异常生成的过氧化氢会引起细胞氧化损伤和一些疾病进程,如哮喘、动脉粥样硬化、骨质疏松和神经退行性病变。

原理
过氧化物检测试剂盒提供简单且高通量的适应性测定法,用于定量测定无需预处理的多种生物样品(如血清、柠檬酸盐-血浆、尿液、细胞裂解液和培养基)中的过氧化物浓度。该检测利用Fe3+ −二甲酚橙显色反应,样品中的过氧化物将Fe2+氧化为Fe3+,(与二甲酚橙)形成可通过比色法(585 nm)测定的紫色复合物,与过氧化物浓度成正比。优化后的配方可减少原始样品中其他物质的干扰。1 μM H2O2的浓度相当于34 ng/mL或34 ppb。该试剂盒的检测范围为0.2–30 μM (7–1,020 ng/mL)H2O2(96孔板形式)。
