一般描述
GAPDH(甘油醛-3-磷酸脱氢酶;EC 1.2.1.12)可催化甘油醛-3-磷酸酯(GAP)转化为1,3-二磷酸甘油酸盐(BPG),对糖酵解具有重要作用。该酶参与了细胞凋亡、膜转运、铁代谢和核移位等细胞过程。GAPDH(管家基因)表达是稳定的组成型表达。GAPDH活性异常与细胞增殖异常和癌变相关。GAPDH活性的准确定量对于疾病诊断和研究正常细胞生理学非常重要。

原理
GAPDH 活性检测试剂盒为监测各种样品中的 GAPDH 活性提供了一种简单而灵敏的方法。GAPDH 的活性是通过偶联酶反应测定的,其中 GAP 被 GAPDH 转化为 BPG。这导致比色(450nm)产物与存在的酶活性成比例。该检测对 100 mUnites/mL 敏感。一个单位的 GAPDH 活性是指在 pH7.2,37℃ 下,每分钟产生 1.0 mmole NADH 的酶量。
