一般描述

脂肪分解是将甘油三酯水解成游离脂肪酸和甘油的过程。 该过程涉及到脂肪TG脂肪酶（ATGL）、激素敏感性脂肪酶（HSL）和甘油单酯脂肪酶的作用。脂肪分解可通过提供氧化代谢的底物来维持禁食和运动期间的能量平衡。脂肪分解受营养因子和激素的调节。脂肪分解作用调节存在的问题与肥胖、糖尿病和代谢综合征相关。

适用性

高灵敏试剂盒可用于检测3T3-L1细胞及脂肪细胞中的脂解作用。

原理

高灵敏度脂解（3T3-L1）荧光法测定试剂盒含有可激活β-肾上腺素能受体的合成儿茶酚胺（或替代肾上腺素）。这导致将ATP转化为cAMP的腺苷酸环化酶激活。然后cAMP通过激素敏感性异常酶激活甘油三酯的沉淀。通过测量与甘油含量成比例的荧光产物（λex = 535/λem = 587 nm）测定脂解。
