一般描述

由于电子传递链的酶活性,使线粒体产生跨膜电位。在发生细胞凋亡时,线粒体膜电位(MMP)消失,线粒体通透性转运孔开放,导致细胞色素c释放到细胞质中,进而引发细胞凋亡级联中的其他下游事件。

应用

线粒体膜电位检测试剂盒已被用于估算中国仓鼠卵巢(CHO)细胞的线粒体膜电位。它已被用于线粒体膜通透性检测(MMP检测) ,还用于检测线粒体膜通透性的消失。

适用性

本试剂盒适于检测哺乳动物细胞中的线粒体跨膜电势差(MMP),适合利用酶标仪筛选凋亡抑制剂及激活剂等。

原理

线粒体膜电位试剂盒经优化适用于检测细胞中MMP消失。阳离子疏水性线粒体膜电位染料由于线粒体膜电位的存在而在正常线粒体中积累,导致荧光增加(λex = 540/λem = 590 nm)。在凋亡细胞中,MMP消失,导致荧光减少。该试剂盒可用于监测细胞凋亡以及筛选凋亡抑制剂和激活剂。
