一般描述
ADP/ATP比例变化已用于区分细胞死亡和活率模式。ATP水平升高、ADP水平降低表示细胞正在增殖。相反,ATP降低、ADP的升高表示细胞凋亡或坏死,且相对于凋亡,坏死细胞ATP降低和ADP升高程度更为明显。

原理
ADP/ATP比率测定试剂盒提供了一种简单直接的细胞ADP和ATP水平测定方法,可用于筛查细胞凋亡、坏死和增殖。该测定包括两个步骤。第一步,利用工作试剂裂解细胞以释放ATP和ADP。在存在萤光素酶的条件下,ATP立即与底物D-萤光素反应产生荧光。光强度是细胞内ATP浓度的直接指标。

ATP + D-萤光素 + O2 ----------> 氧化萤光素 + AMP + PPi + CO2 + 荧光
第二步,ADP通过酶解反应转化为ATP。然后像第一步一样,新形成的ATP与D-荧光素反应。测得的第二个光强度代表样品中的总ADP和ATP浓度。
