应用
葡萄糖摄取检测试剂盒(荧光法)已可用于测量单核细胞样髓源抑制细胞(M-MDSC)的葡萄糖摄取量。
适用性
适用于检测在96孔微量滴定板中培养的贴壁细胞或悬浮细胞的葡萄糖摄取。
原理
葡萄糖摄取检测试剂盒(荧光法)可简单、直接地测定多种细胞的葡萄糖摄取能力。该试剂盒使用葡萄糖类似物2-脱氧葡萄糖(2-DG)测量葡萄糖摄取,2-DG在测定中被细胞摄取并被己糖激酶磷酸化为2-DG6P。2-DG6P无法进入后续代谢步骤,因而会在细胞中聚集,直接成比例地反映细胞的葡萄糖摄取水平。在该测定中,2-DG的摄取水平通过偶联酶促反应方法测定,其中,2-DG6P在此过程中被氧化生成NADPH,随后NADPH与探针反应,生成的荧光(λex = 535/λem = 587 nm)产物与细胞的2-DG摄取量成比例。
