一般描述
血流中D-乳酸的浓度通常是纳摩尔级,由肠源或甲基乙二醛途径产生。在哺乳动物中,D-乳酸代谢需要D-乳酸氢化酶且代谢缓慢,因此血液浓度水平增加可导致酸血症和酸中毒。D-乳酸酸中毒的严重程度可能与神经毒性症状有关。体内D-乳酸显著积累也可能与代谢和排泄受损有关。
应用
D-乳酸比色检测试剂盒用于检测产生的乳酸的立体特异性。
适用性
适用于血清、血浆、细胞、培养物和发酵培养基样品。
原理
在该检测中,D-乳酸被D-乳酸氢化酶特异性氧化,产生与之成比例、可在450 nm处进行检测的比色产物。样品中D-乳酸的有效浓度范围是0.1-10 mM。
