一般描述

MAP激酶激酶(MAPKK、MEK1和MEK2)是对苏氨酸/酪氨酸残基双磷酸化激活底物的酶家族成员。MEK1和MEK2的催化结构域有86%的同源性。MEK1/2先被丝裂原刺激细胞中的Raf-1激酶活化, MEK1/2随后激活ERK1/2,进而激活多个下游靶标,参与细胞增殖、凋亡和移动。除了ERK1/2,可能还有其他非催化性的MEK1/2效应子。MEK的各种亚型在中枢神经系统、胸腺、脾脏、心脏、肺和肾脏中广泛表达。 活性MEK1/2是NIH3T3细胞转化和PC-12细胞分化的充分条件。
该抗体仅特异性识别Ser217和Ser221位磷酸化的MEK1/2 (45 kDa)。它与仅Ser217为磷酸化的MEK1/2反应强烈,但与仅Ser221磷酸化的MEK1/2反应较弱。与其他相关家族成员(包括SEK)没有交叉反应。

应用

抗-磷酸MAP激酶激酶1/2(MEK1/2)抗体应用广泛。对于血清处理的NIH3T3细胞的免疫印迹分析,推荐的工作稀释度为1:1000。对于免疫沉淀,可使用1:50的工作稀释度。对于石蜡包埋的人乳腺癌切片的免疫组织化学检测,可使用1:100的抗体稀释液。对于HeLa细胞的免疫细胞化学应用(免疫荧光),可使用1:1000的工作稀释度。

10 mM HEPES、pH 7.5溶液,含有150 mM氯化钠、100 μg/ml牛血清白蛋白和50%甘油。
