层粘连蛋白是一种普遍存在的非胶原结缔组织糖蛋白,是基底膜的主要成分。层粘连蛋白是细胞外基质蛋白。LAMA1(层粘连蛋白亚基α1)基因位于人染色体18p11上。通过免疫特异性纯化方法从抗血清中分离抗体。抗原特异性分离可去除几乎所有兔血清蛋白,包括不与层粘连蛋白特异性结合的免疫球蛋白。

特异性

通过对福尔马林固定石蜡包埋的人或动物组织切片进行间接免疫荧光标记和斑点杂交免疫测定,确定抗层粘连蛋白抗体的特异性。 通过间接免疫荧光分析发现,抗体可以对未被酶促反应掩蔽的人和动物组织进行特异性基底膜染色。 在斑点杂交印迹免疫测定中,兔抗层粘连蛋白抗体与层粘连蛋白反应,但不与纤连蛋白、玻连蛋白、IV型胶原蛋白或A、B和C型硫酸软骨素反应。层粘连蛋白的亲和分离抗体与人、哺乳动物、禽类、爬行动物和两栖动物来源的层粘连蛋白反应。

免疫原

层粘连蛋白是从小鼠Engelbreth-Holm-Swarm(EHS)肉瘤的基底膜中分离得到的。

应用

兔抗层粘连蛋白抗体可用于免疫组织化学以标记不同物种的血管壁、基底膜相关各种疾病过程的分类、人类肿瘤起源的鉴定及其分类以及区分非侵入性和侵入性病变。该抗体可用于监测生物体液中的层粘连蛋白水平和体内基底膜病变的实验性产生。
使用50 ng/dot的层粘连蛋白进行斑点杂交印迹免疫测定,确定抗体的工作稀释度至少为1:1,000。
使用福尔马林固定石蜡包埋的人和动物组织进行间接免疫组织学分析,确定抗体的工作稀释度至少为1:25。
抗层粘连蛋白抗体可用于免疫组织化学染色 免疫组织化学分析。

生化/生理作用

LAMA1(层粘连蛋白亚基α1)参与多种疾病,如癌症、感染、炎症性疾病和自身免疫性疾病。LAMA1突变可导致小脑发育不良以及有或无视网膜营养不良的囊肿。LAMA1在基底膜组装和胚胎早期生长中也是至关重要的。

外形

溶于0.01M磷酸盐缓冲盐水,pH 7.4,含有1% BSA和15 mM 叠氮化钠。

储存及稳定性

若需连续使用,可在2-8°C下保存最长一个月。若需延长储存时间,可将溶液分装并冷冻。不建议反复冻融或存放在“无霜”冰柜中。如果长期储存时出现轻微浑浊,请在使用前通过离心澄清溶液。
