一般描述
凝集素是一类普遍存在于真菌、植物和动物体内的碳水化合物结合蛋白。植物和动物中对凝集素的结构研究较多。该蛋白的二级结构富含 β-链,并在表面拥有一个碳水化合物结合部位。
应用
来自 美洲商陆的凝集素已被用于:
- 作为单核细胞培养的培养基补充物,以诱导淋巴细胞增殖
- 作为阳性对照研究趋化因子(C-C基序)配体2(CCL2)在蜕膜白细胞增殖和凋亡中的作用
- 作为RPMI(洛斯维·帕克纪念研究所)1640号培养基的补充剂,用于外周血淋巴细胞培养
生化/生理作用
凝集素在糖缀合物的表征、成像和靶向性方面具有重要作用。它在微阵列分析中的应用,使有效的糖组分析成为可能。这是因为其与寡糖、糖蛋白和糖脂的特异性相互作用。在植物和真菌中,凝集素防御病原体/食料。凝集素参与宿主的识别和组织的黏附,参与微生物的致病过程。
PWM 从商陆根中提取。该蛋白质具有红血球凝集、白血球凝集和促有丝分裂特性。PWM 不具有血型特异性,但具有与 N-乙酰基-β-D-葡糖胺低聚物的亲和性。
