一般描述

本品为从鼠伤寒沙门菌中提取的苯酚。来源菌株为 ATCC 7823。该 LPS 已被用于诱导大鼠和豚鼠的 NOS。

应用

脂多糖(LPS)是革兰氏阴性细菌细胞壁的特征组分。LPS及其脂质A部分通过Toll样受体4(TLR4)刺激先天免疫系统的细胞,Toll样受体 4是Toll样受体蛋白家族的成员,其识别常见的病原体相关分子模式(PAMP)。

生化/生理作用

脂多糖 (LPS) 定位于膜的外层,在无荚膜的菌株中暴露于细胞表面。它们有助于外膜的完整性,并保护细胞免受胆盐和亲脂性抗生素的操作。

制备说明

本品可溶于水 (5 mg/mL) 或细胞培养基 (1 mg/mL),得到淡黄色浑浊溶液。涡旋并加热至 70-80 oC 后,在盐水中获得了更浓缩但仍浑浊的溶液 (20 mg/mL)。脂多糖是在每种溶剂中形成胶束的分子。在水和磷酸盐缓冲盐水中观察到浑浊溶液。有机溶剂不能提供更清晰的溶液。甲醇产生浑浊的带有悬浮物的混悬液,而水产生均匀的浑浊溶液。
