应用

通用蛋白质印迹方案:
  • 糖蛋白样品规格:500ng
  • 凝集素浓度: 0.1ug/ml

  1. 每条泳道上样500 ng糖蛋白样品
  2. 进行4-20% Bis-Tris SDS page凝胶
  3. 转膜凝胶至PVDF膜上
  4. 用于RIPA缓冲液(R0278 Sigma)在室温对膜进行封闭1小时

生化/生理作用

BS-I对末端α-D-半乳糖残基具有主要亲和力,对末端N-乙酰基-α-D-半乳糖胺残基具有二级亲和力。

BS-I是一种四聚体凝集素,由两种类型的亚基A和B组成。存在由不同亚基组成的五种BS-I同工凝集素:BSI-B4、BSI-AB3、BSI-A2B2、BSI-A3B、以及BSI-A4。BSI-B4 是血型B特异的,其对末端α-D-半乳糖残基具有独特的亲和力,而BSI-A4 具有血型A特异性,其对末端N-乙酰基-α-D-半乳糖胺残基具有主要亲和力。

分析说明

凝集活性以 μg/mL表示,并且使用磷酸盐缓冲盐水(pH6.8)对1mg/mL溶液进行连续稀释液来测定,对于每种凝集素,所含有钙、镁和锰的浓度不同。该活性是在25℃孵育1小时后,对2%合适的红细胞悬浮液进行凝集所需的最低浓度。
