Biochem/physiol Actions
Glucosaminyl (N-acetyl) transferase 2 (GCNT2) is an enzyme essential for the development of the blood group I antigen. Biallelic mutation of GCNT2 leads to autosomal recessive congenital cataracts and the rare adult i blood group. GCNT2 associates with β1,3-N-acetylglucosminyltransferase 2(β3GnT2) to regulate the PLN levels in olfactory neurons by modulating β1, 6-branches that aids in Poly-N-acetyllactosamine (PLN) extension. Over human developmental stage, GCNT2 enzyme transforms linear i-antigen to I-branching glycan by replacing GlcNAc of uridine diphosphate–GlcNAc with galactose β1-6 linkage of linear lactosamine chains. Uprooted expression of GCNT2 enzyme increases cell detachment, cell adhesion, cell migration and invasion in vitro, and lung metastasis of breast cancer cells in vivo. Overexpression of GCNT2 contributes to breast cancer metastasis. Transforming growth factor-β1 (TGF-β1) acts as an effector of GCNT2 enzyme and plays a major role in TGF-β1induced epithelial-to-mesenchymal transition (EMT).

General description
Glucosaminyl (N-acetyl) transferase 2 (GCNT2) protein is encoded by the novel metastasis-related gene mapped to human chromosome 6p24. The GCNT2 protein belongs to the β-1, 6-N-acetylglucosaminyltransferase family, which has an essential role in glycosylation.
