7,8-dihydro-8-oxoguanine triphosphatase (NUDT1) is a hydrolase belonging to the Nudix family. The gene encoding it is located on chromosome 7p22. NUDT1 is localized in the cytoplasm and mitochondria. It is expressed in the human thymus, testis and embryonic tissues. NUDT1 has a conserved Nudix motif and a substrate-binding pocket-forming helix.

生化/生理作用

7,8-dihydro-8-oxoguanine triphosphatase (NUDT1) hydrolyzes the oxidized forms of dATP and dGTP like 2-hydroxy-dATP (2-OH-dATP), 8-hydroxy-dATP (8-OH-dATP) and 8-dihydro-8-oxo-dGTP (8-oxo-dGTP) to the corresponding monophosphates. By doing so, it prevents the misincorporation of the oxidized purine nucleoside triphosphates into DNA and in turn inhibits A:T to C:G and G:C to T:A transversions from occurring. It prevents the formation of various types of tumors.
