粒细胞集落刺激因子(G-CSF)是受限嗜中性粒细胞系的造血细胞的增殖、分化、存活和激活因子。它是由被内毒素(LPS)激活的巨噬细胞、被TNF-α和IFNγ激活的单核细胞、被IL-1或TNF-α945;激活的成纤维细胞和内皮细胞以及被IL-1或LPS激活的骨髓基质细胞产生的。这表示,在炎症过程中,内毒素刺激组织巨噬细胞产生G-CSF,同时还产生多种其他细胞因子,包括IL-1和TNF-α,这些细胞因子进一步刺激内皮细胞和成纤维细胞释放更多的G-CSF。G-CSF还可以与IL-3发挥协同作用,缩短早期造血祖细胞的G0期。除了同名的增殖活性外,G-CSF还作用于成熟的中性粒细胞,以提高其存活率并刺激其肿瘤杀伤活性。人类G-CSF结合并激活130 kD至150 kD的糖蛋白单链受体(已被归类为造血(细胞因子)受体家族)、细胞因子I类或gp130相关细胞因子受体家族(尽管它显然不与gp130结合)。G-CSF受体存在于嗜中性粒细胞、响应G-CSF的髓样白血病细胞、嗜中性粒细胞系的骨髓细胞以及胎盘滋养层中,并且可以表达为可溶形式。通过可变剪接能够从单个基因合成两种形式的人G-CSF(177和174个氨基酸),但是鼠G-CSF只能表达为178个氨基酸这一种形式。人和鼠G-CSF具有73%的氨基酸序列同源性和完全交叉反应性。
