MITF (microphthalmia-associated transcription factor) is a b-HLH-Zip (basic helix-loop-helix leucine zipper) transcription factor, which is specific to melanocytes. It is a member of the Myc superfamily, and the gene is localized to human chromosome 3. It is 229kb in size. It belongs to the MiT (microphthalmia) family of transcription factors, which also includes TFEB (transcription factor EB), TFEC and TFE3. Due to alternative splicing, this protein is produced as multiple different isoforms such as TF-A, MITF-B, MITF-C, MITF-D, MITF-E, MITF-H, MITF-J, MITF-M and MITF-Mc. The N-termini of these isoforms vary, and they have different tissue expression patterns. The shortest isoform is MITF-M, which is only expressed in melanoma cells and melanocytes. Its transcriptional activity is modulated by its two TADs (transactivation domains), present at N- and C-termini.

应用

Anti-MITF antibody produced in chicken is suitable for western blotting analysis at a dilution of 1:500, for tissue or cell staining at a dilution of 1:200.

生化/生理作用

Microphthalmia-associated transcription factor (MITF) is involved in lineage-specific pathway regulation of many types of cells. Mitf is required for the proper development of osteoclasts, melanocytes, retinal pigment epithelial cells, mast cells and natural killer cells. It can be a valuable adjunct in the diagnosis of metastatic tumors that are suspicious for melanoma but negative for other melanoma markers.
