一般描述

右旋糖酐是一种无水葡萄糖聚合物。由约 95% 的 α-D-(166) 键组成。其余 (163) 连接可解释右旋糖酐的分支。分支长度的冲突数据表明平均分支长度小于 3 个葡萄糖单位。然而,其他研究表明存在超过 50 个葡萄糖单位的分支。已发现天然右旋糖酐分子量 (MW) 在 900 万 ~5 亿范围内。较低分子量的右旋糖酐将表现出略少的分支,并且分子量分布范围更窄。分子量大于 10,000 的右旋糖酐表现出高度支链状。随着分子量的增加,葡聚糖分子的对称性增强。分子量为2,000至10,000的葡聚糖表现出可膨胀线圈的特性。当分子量低于 2000 时,右旋糖酐更像棒状。采用以下一种或多种方法测定右旋糖酐的分子量:低角度激光散射、尺寸排阻色谱、铜络合和蒽酮试剂比色法还原糖测定和粘度。

应用

FITC-右旋糖酐作为荧光通量示踪化合物用于心血管、微循环、灌注、细胞单层和细胞膜渗透性研究,支持血流、膜损伤、血管引流和肾脏消除等过程的测量。小FITC-右旋糖酐也用于测量内吞作用和细胞连接通透性等过程。500 kDa 荧光素-右旋糖酐可用作脑脊液 (CSF) 示踪剂。FITC-右旋糖酐-500 可用于研究内皮细胞或上皮细胞单层孔隙度、细胞层内顶端基底侧运动和细胞连接渗透性。
FITC-右旋糖酐广泛用于利用微荧光法的微循环和细胞渗透性研究。FITC-右旋糖酐已被用于研究植物细胞壁孔隙率和毛细管渗透性。已证实血浆蛋白不与 FITC-右旋糖酐结合。
用异硫氰酸荧光素标记的葡聚糖可用于动物的灌注研究。

制备说明

Sigma通常以25mg / ml或更高的浓度测试FITC右旋糖酐在水中的溶解度。溶液应避光保存。在体内,FITC-右旋糖酐可稳定 24 小时以上。
