一般描述

纤连蛋白是一种具有多个结构域的糖蛋白。它包含一个N端、中央结合域和一个肝素结合域。牛血浆纤连蛋白具有带二硫键的多肽链，分子量分别为220和215 kDa。
纤连蛋白是粘着糖蛋白，存在于细胞表面和血浆中。纤连蛋白（FN）是一种多功能糖蛋白，相应的基因定位于人染色体2q34-36。该基因有75kb长并含有50个外显子。它存在有两种主要的异构体，一个可溶而另一种不可溶。前者存在与血浆中，而不可溶形式则存在于组织和软骨的细胞外基质（ECM）中。因在不同的区域存在可变剪切，故存在约20种不同的FN形式，其中有两个III型外显子的被称为Extra Domains A和B。

应用

来自于牛血浆的纤连蛋白可用于以下应用：
  • 用于涂布玻璃盖玻片以用于蜕膜基质细胞（DEC）的培养
  • 细胞运动检测（培养板涂有纤连蛋白）
  • 细胞扩散和免疫荧光

生化/生理作用

纤连蛋白是蛋白质支架的一种调节因子。它协调细胞过程，如血小板功能、伤口愈合、粘附、迁移和聚集。研究表明人牙髓 (HDP) 细胞依靠纤连蛋白 (FN)实现最初的附着和之后的扩散。FN可能具有促有丝分裂活性，促进细胞增殖，也在髓细胞向成牙质细胞的分化过程的调节中起一定作用。FN 的存在影响 HDP细胞的发展和命运，特别是在分化为成牙质细胞的过程中，成牙质细胞可形成牙齿中的牙本质。
