一般描述
小鼠抗因子IX单克隆抗体（小鼠IgG1同种型）来源于小鼠Sp2/0-Ag14骨髓瘤细胞与人血浆纯化因子IX免疫BALB/c小鼠脾细胞融合产生的HIN-1杂交瘤。人血浆中因子IX的浓度范围为2.5-5 mg/mL，其半衰期约为24小时。人因子IX基因的大小约为40 Kb，位于X染色体的远端。
特异性
单克隆抗因子IX是一种不依赖二价阳离子的抗体，当用于未变性、未还原的人血浆的免疫印迹时，可识别因子IX。它也可用作与单克隆抗因子IX克隆HIX-5（产品编号F1020）的夹心型免疫测定中的配对标记抗体。
该抗体可用于纯化因子IX，并制备去除因子IX的人血浆。

应用

生化/生理作用
FIX在内源性和外源性凝血系统中起关键作用。它在肝脏中合成，随后在血浆中活化后转化为丝氨酸蛋白酶。在第二步中，FIX经历一系列复杂的翻译后修饰，例如12个N-末端谷氨酸残基的γ-羧化，N-和O-连接的糖基化，磷酸化，β-羟基化，硫酸化，二硫键形成等，翻译后修饰完成后分泌到血浆中。因子IX的遗传性缺陷或功能障碍会导致B型血友病或“圣诞节病”（所描述的第一个家族的姓氏）。在血友病B中，FIX显示凝血功能受损和出血倾向增加，这是X染色体连锁FIX基因突变的原因。因子IX在肝实质细胞中合成，需要翻译后维生素K依赖性修饰才能成为成熟的血浆酶原。当患者缺乏维生素K或服用干扰维生素K代谢的口服抗凝剂时，会诱发低凝或抗血栓形成状态。
