一般描述
肠激酶也称为肠肽酶,是一种跨膜蛋白质。这种肠蛋白酶由一条重链和1一条轻链通过二硫键连接而成。牛肠肽酶的氨基端序列类似于胰蛋白酶样丝氨酸蛋白酶。肠激酶是一种高度特异性丝氨酸蛋白酶,可用于去除N-末端和Met-N-末端融合蛋白中的FLAG多肽,但不去除C端FLAG。
应用
牛肠激酶已用于去除重组谷胱甘肽过氧化物酶1(Gpx1)中的S标签和N端His标签。它还用于切除(聚羟基脂肪酸酯)颗粒中的白介素-2(IL-2)蛋白。
肠激酶是 S1 肽酶家族的一员。在体内,它负责胰蛋白酶原胰蛋白酶的蛋白水解活化。肠激酶用于位点含有可进入肠激酶识别位点的重组融合蛋白进行位点特异性切割,以去除亲和标签。
生化/生理作用
