在存在抗氧化剂的条件下，自由基阳离子受抑制的程度取决于抗氧化剂活性，且颜色深度成比例下降。Trolox是一种水溶性维生素E类似物，可作为标准抗氧化剂或对照抗氧化剂。
自由基或活性氧(ROS)在生化氧化还原反应过程产生，是正常生理细胞新陈代谢（免受传染性生物体侵害）的参与成分，并可对紫外线、香烟烟雾、环境污染物和伽马辐射等环境因素作出响应。ROS形成之后即会攻击细胞成分，破坏脂质、蛋白质和DNA，从而引发多种疾病，包括癌症、动脉粥样硬化、类风湿性关节炎、糖尿病、肝损伤和中枢神经系统疾病。

应用

它已在A431和CHO细胞裂解物，大鼠脑、肝和肾裂解物，人血浆、血清、尿液和唾液中进行了测试。
抗氧化测定试剂盒提供了有效测定血浆、血清、尿液、唾液、细胞和组织裂解液的总抗氧化能力所需的所有试剂。它也被用来测定自由基阳离子（2,2′-连氮基-二-[3-乙基苯并噻唑啉磺酸盐]）（ABTS +）的清除能力。

原理

抗氧化剂测定的原理是：由肌红蛋白和过氧化氢形成亚铁肌红蛋白自由基，该自由基将ABTS（2,2¢-联氮双(3-乙基苯并噻唑啉-6-磺酸）氧化生成自由基阳离子ABTS·+（绿色可溶性色原），从而在405 nm处用分光光度法测定。

制备说明

试剂用超纯水配置。

重悬

肌红蛋白溶液保存在-20℃条件下。
Trolox工作溶液保存在-20℃条件下。
ABTS底物溶液可在4℃下保存两周或-20℃下保存1个月。
