General description

抗X型胶原蛋白单克隆抗体(小鼠IgM同种型)源自COL-10杂交瘤,该杂交瘤是通过将小鼠骨髓瘤细胞与用X型猪胶原蛋白免疫的BALB/c小鼠的脾细胞融合而产生的。X型胶原蛋白具有相似的结构域结构与VIII型胶原蛋白:50 kDa的中央三螺旋(COL1)结构域两侧是N端(NC2)和C端(NC1)非三螺旋结构域。
胶原蛋白是由三种多肽组成的细胞外糖蛋白,它们混合形成三重螺旋。X型胶原蛋白是在长骨,椎骨和肋骨生长区的胎儿肥大软骨中发现的59 kDα1(X)链的同源三聚体,而在成年人中,它也存在于甲状软骨中。单克隆抗胶原X型抗体可用于细胞培养物的组织学和免疫组织化学评估。它也可用于研究特定的差异组织表达。

Biochem/physiol Actions

X型胶原是肥大软骨细胞的产物。X型胶原是非纤维状的,但形成与软骨胶原有关的细的细胞周细丝。它与基质蛋白(如连接蛋白V、软骨钙黄绿素、胶原II和蛋白聚糖)以及Ca2+相互作用。它在软骨内骨化过程中充当避免局部衰竭的支架,并且在软骨矿化中也起作用。人α1(X)胶原基因的NC1编码域中的突变与Schmid干骺端软骨形成有关。
