一般描述
针对蛋白质的定制生物素化，Sigma 提供了两种不同规模的偶联试剂盒。当需要高灵敏度和特异性时，生物素-抗生物素蛋白系统受到青睐。生物素可以与抗体、凝集素、酶和其他蛋白质结合。 试剂盒中的操作方案已针对抗体进行了优化。抗生物素蛋白以高亲和力（Ka= 1015 M）和特异性与生物素结合。当生物素蛋白与酶或荧光染料结合后，可通过酶促反应将底物底物转化为形成可见产物，或通过分光光度法或流式细胞术检测荧光来鉴定生物素化化合物。
应用
ImmunoProbe™生物素化试剂盒用于制备生物素标记抗体。
生物素已用氨基己酸酯修饰，然后通过与磺基-N-羟基琥珀酰亚胺 （BAC-Sulfo-NHS）的酯键活化。氨基己酸酯提供了一个六碳间隔子，可减少生物素上的空间位阻并提高亲和素上结合位点的可及性。羟基琥珀酰亚胺的磺化增加了试剂的极性，使其在水性缓冲液中易于溶解。该酯提供了与不稳定酯键相邻的羰基碳作为可及性赖氨酸残基的伯胺侧链的靶标，通过酰胺键将生物素氨基己酸酯与蛋白质连接。
