5-溴-4-氯-3-吲哚基磷酸酯 (BCIP)/硝基蓝四唑(NBT)/硝基蓝四唑鎓(NBT)是用于检测碱性磷酸酶(AP)的显色底物系统。AP水解BCIP形成中间体无色靛蓝化合物,并进一步被NBT氧化形成5,5′-二溴-4,4′-二氯靛蓝(BCI)。其中,NBT被还原为不溶的紫色双甲双胍(DF),形成NBT-DF。NBT-DF和BCI的混合物形成深紫色沉淀。AP可水解多种BCIP分子,因此成为了一种高灵敏度方法。
