一般描述
L-氨基酸氧化酶(LAAO)是一种黄素蛋白,分子量为130 kDa。它由两个大约70 kDa的不同亚基组成。全酶的每个分子都有两个黄素腺嘌呤二核苷酸(FAD)分子。LAAO是一种糖蛋白,含有大约2-5%的碳水化合物,包括唾液酸。最适pH约为7.5。除微生物和动物组织外,它还存在于许多蛇毒液中,尤其是在肾脏中。在N-末端区域,它有一个带谷氨酸残基的βαβ结构域。LAAO使毒液呈黄色。
应用
来自响尾蛇的L-氨基酸氧化酶已被用于定量测定亚胺脱氨酶活性中的rid 蛋白质。
L-氨基酸氧化酶 (LAAO) 可用于将L-氨基酸转化为相应的α-酮酸。来自Sigma的L-氨基酸氧化酶已被用于亮氨酸氨基肽酶(LAP)活性测定。该酶为固定化酶,可用于化学发光检测酶促流动注射方法中进行L-丙氨酸的现场测定。
生化/生理作用
来自响尾蛇的L-氨基酸氧化酶(LAAO)需要Mg2 +才能激活。N末端区域对于FAD的结合是必不可少的。通过LAAO的氧化反应生成的 H2O2可防止血小板聚集,并引起水肿和细胞凋亡。乙二胺四乙酸(EDTA)和苯甲基磺酰氟(PMSF)可抑制LAAO的活性。
