一般描述
亲和素是一种同源四聚体糖蛋白,存在于鸟类、爬行动物和两栖动物的蛋清中。每个亚基为16 kDa,单糖基化并以更大的亲和力和特异性与生物素分子结合。在等电点(pI)和抗原特性(除了分子量)方面,玉米来源的重组抗生物素蛋白与鸡蛋来源的抗生物素蛋白相似。玉米来源的抗生物素蛋白比鸡蛋来源的抗生物素蛋白的分子量低。这可能是由于植物中糖基化组成较不复杂。从玉米中商业化生产抗生物素蛋白在可获得更大的生物量和避免动物病毒的共纯化方面具有一定的优势。
应用
亲和素与d-生物素(Kd ~ 10-15)形成极强的复合物。这种活性使抗生物素蛋白和生物素在检测和定量的免疫化学方法中都非常有用。
