General description

β-淀粉样蛋白前体蛋白 (APP) 是由蛋白水解酶β-分泌酶 (BACE1) 和 γ-分泌酶依次裂解,生成以Aβ1-42和 Aβ1-40型为主的β-淀粉样蛋白 (Aβ) 肽。分泌的Aβ肽通过再吸收机制和之后的内体降解过程降解,或通过胞外胰岛素降解酶降解。Aβ的胞外积累导致聚集体和原纤维的形成,最终形成淀粉样蛋白沉着物神经炎斑,其是阿尔茨海默病 (AD) 的标志物。
淀粉样β-肽(Aβ), 一种 含38至 43 个氨基酸的肽,来源于β-淀粉样前体蛋白(APP)。β-淀粉样蛋白基因位于人染色体21q21上。APP是一种I型跨膜蛋白。它在内质网(ER)中产生,然后在高尔基体的协助下迁移至反面高尔基网(trans-Golgi-network,TGN)。

该抗体可识别人 β-淀粉样肽、全长淀粉样前体蛋白 (APP)、可溶性 APP(sAPPβ′和 sAPPα)、C99 裂解 形式和 Aβ(1-40/42),但不是可溶性 APP 形式 sAPPβ。
