一般描述

二氢罗丹明123(DHR123)是一种不带电荷的非荧光染料,是罗丹明123(R123)衍生物。存在活性氧物质的情况下,细胞内的二氢罗丹明123(DHR123)氧化为荧光R123,定位在线粒体中。[1]它是2,7-二氯二氢荧光素(DCDHF)的类似物,具有亲脂性,通过细胞膜扩散。[2]

应用

二氢罗丹明123用于检测头肾来源巨噬细胞的细胞内自由基或ROS(活性氧物质)。[3]还用于红细胞DHR测定[4]和中性粒细胞氧化爆发测定。 [5]

生化/生理作用

二氢罗丹明123可检测单细胞水平的超氧离子[6] 可用于在呼吸爆发事件中探测中性粒细胞超氧化物的产生。[1]也用于检测巨噬细胞[2]和人类精子的氧化应激情况。 [1]
二氢罗丹明123可用于检测ROS(活性氧物质)。ROS的存在可将二氢罗丹明123氧化成荧光衍生物罗丹明123。该反应可在过氧化物酶或类似催化剂存在下发生。[7][8]
