General description

使用AMV RT进行第一链cDNA合成具有一定优势。与M-MuLV RT相比，AMV RT具有更高的热稳定性，与在+ 37°C进行的反应相比，在+ 42°C下进行的反应，具有更高的特异性和更好的二级结构解析度。

扩增RNA需要将RNA底物转化为DNA。这一过程可以通过使用逆转录酶实现，例如使用AMV RT（禽成髓细胞瘤病毒逆转录酶）或M-MuLV RT（莫洛尼氏鼠白血病病毒逆转录酶）。所得cDNA可用作标准PCR模板。
AMV RT在一个或多个位点合成新的cDNA链，而位点由所用引物的类型决定：

如使用Oligo-p(dT)₁₅用作引物，则位点在聚(A)mRNA的3′端
如使用随机引物p（dN）₆，则位点在沿着mRNA模板的非特异性点处，或
在由序列特异性引物确定的位点处。

第一链cDNA合成试剂盒以两步RT PCR用作起始反应，合成第一链cDNA。本试剂盒包含用于第一链合成的逆转录酶AMV、两种不同引物、PCR核苷酸混合物和对照 Neo pa RNA。可以使用标准程序克隆RT-PCR生成的PCR产物。
