抗地高辛-荧光素，Fab片段带有来自多克隆抗地高辛抗体的fab片段，并且与荧光素缀合。
特异性
来自绵羊的多克隆抗体对地高辛配基和地高辛具有特异性，并且与其他类固醇（如人类雌激素和雄激素）没有交叉反应性。
应用
抗地高辛-荧光素，Fab片段可用于检测地高辛标记的化合物：
- 糖结合物研究中地高辛标记的糖
- 荧光 原位 杂交（FISH）
- 免疫组织细胞化学
- 原位 杂交
制备说明
- 工作浓度：结合物的工作浓度取决于应用类型和底物。以下浓度可作为指导：检测糖蛋白中地高辛标记的糖：50至20μg/ml
- 荧光原位 杂交(FISH)：1至20μg/ml
- 免疫组织细胞化学：20至50μg/ml
- 原位 杂交：20至50μg/ml
抗DIG抗体具有高度特异性和敏感性。因此，对于FISH，1μg/ml的浓度已足够，并且仅在关键探针的情况下，该浓度可以升高至<10μg/ml（可能产生明显的背景信号）。
工作溶液：溶解
加入1ml双蒸水至终浓度为200μg/ml。
封闭：
PBS，0.5％牛血清白蛋白(w/v)，pH7.4。
1％封闭试剂(w/v)，1至5％热灭活的胎牛血清(v/v)或绵羊正常血清（可用于减少非特异性结合）。此外，pH可以增加至pH8.5至9.0。
储存条件（工作溶液）：每次请制备新鲜溶液！
用地高辛免疫后，通过离子交换层析纯化绵羊IgG，并通过免疫吸附分离特异性IgG。通过木瓜蛋白酶消化获得的Fab片段，可通过凝胶过滤进行纯化，并与特异性标记缀合，并稳定于缓冲液中。
重悬
加入1ml双蒸水至终浓度为200μg/ml。
