一般描述

转化生长因子β(TGFβ)系统属于细胞因子超家族和信号通路成员,在动物界进化上高度保守。TGFβ参与免疫细胞发育、血管生成、伤口愈合、胚胎发育和骨发育过程,在免疫应答、心脏病和肿瘤中起重要作用。正常状态的TGFβ是少数几种能够抑制细胞生长的蛋白,通过使有丝分裂停滞在G1期,促进细胞分化和凋亡。但是,TGFβ信号通路的癌变会使肿瘤细胞能够抵抗TGFβ的正常发挥作用,导致失去调控的增殖发生。初步研究表明,VE-钙粘蛋白可能增强突变的TGFβ信号通路。

MILLIPLEX® TGFβ1单重磁珠检测试剂盒是单因子分析试剂盒,用于定量分析血清、血浆、组织/细胞裂解物和培养上清液中的TGFβ

应用

  • 分析物:TGFβ1
  • 建议种属:除了人类TGFβ1,该试剂盒还可以检测下列动物血清中的TGFβ1:马、兔、小鼠、大鼠、小型猪、犬、仓鼠、豚鼠、恒河猴和食蟹猴。但是,准确的交叉反应量并未测定。
  • 建议的样本类型:血清、血浆、细胞/组织培养物上清液和裂解物
  • 注:本品需要采用提供的试剂和方法对样品进行酸化处理。
  • 建议的样本稀释方法:血清和血浆样本,在酸化处理后1:30稀释,每孔加样25 μL;细胞/组织培养样本,需要在酸化前稀释(详见说明书)。
  • 检测耗时:在2-8℃下过夜(16-18小时)或在室温(20-25℃)下2小时。
