一般描述

可在单个细胞水平实现细胞凋亡的检测。易于检测凋亡形态。通过流式细胞术或荧光显微镜进行检测。试剂盒固封剂可观察总细胞数量。
货号QIA39是我们已被广泛引用的荧光素偶联版本的TdT Colorimetric FragEL DNA片段检测试剂盒的(货号QIA33),可用于非同位素标记。末端脱氧核苷酸转移酶(TdT)可结合响应凋亡信号而产生的DNA片段的暴露3′-OH末端,并催化生物素标记的和未标记的脱氧核苷酸加成。当激发时,荧光素产生强烈的信号,该信号可以通过荧光显微镜或流式细胞仪进行检测。固封剂可维持来自载玻片上标记样品的荧光信号,因此有助于正常细胞和凋亡细胞的形态评估和表征。非凋亡细胞可以通过DAPI滤镜进行观察。

原理

Calbiochem Fluorescein-FragEL DNA碎片检测试剂盒是一种非同位素系统,可用于标记石蜡包埋的组织切片、组织冷冻切片、固定在载玻片上的细胞制剂、或细胞悬液中凋亡核中的DNA断裂。标记的检测和分析可以通过流式细胞仪或荧光显微镜进行。

储存及稳定性

荧光素-FragEL 试剂盒组分在干冰上进行运输。收到后,将试剂盒存放在-20°C非防冻冰箱中。为避免试剂损失在管盖中,请短暂离心所有解冻后的溶液,再取下管盖。(请参见下面的一般注意事项)。
