一般描述
泛素（Ub）最初生产形式为229-a.a.聚泛素-B（UniProt P0CG47）前体蛋白，由人的UBB基因（基因ID 7314）编码或685-a.a.聚泛素C前体蛋白（UniProt P0CG48）由人的UBC基因（基因ID 7316）编码。翻译后去除聚泛素B（Cys229）和聚泛素C（Val685）的C末端，然后进行额外的翻译后裂解，以产生多个拷贝的相同的76个氨基酸的泛素（也称为高迁移率组非组蛋白蛋白HMG-20）分子（每个聚泛素B三个拷贝，每个聚泛素C九个拷贝）。Ub参与细胞蛋白最常见的翻译后修饰(PTM)之一，其中Ub通过其羧基末端（Gly76）与靶蛋白中通常的Lys残基共价连接。给定的目标Lys残基可以连接到一个单个Ub分子（单泛素化）或连接到Ub分子链（多泛素化）。在polyUb链中，Ub分子可以通过七个Lys残基之一（K6、K11、K27、K29、K33、K48和K63）或通过Ub N末端Met1残基（产生线性链）连接。具有单一连接类型的Ub链称为同型，而异型链在同一链中包含混合连接，这可能导致分支/分叉（由于一个Ub连接到两个或多个Ub）或非分支Ub链。Lys48连接的链标记蛋白质的蛋白酶体降解，而Lys63连接的链调节细胞蛋白质的活性/功能。
