General description
NAD(+) hydrolase SARM1 (UniProt: Q6PDS3; also known as EC:3.2.2.6, NADase SARM1, NADP(+) hydrolase SARM1, Sterile alpha and TIR motif-containing protein 1) is encoded by the Sarm1 (also known as Kiaa0524) gene (Gene ID: 237868) in murine species. The encoded protein belongs to the MyD88 (myeloid differentiation primary response gene 88) family of adaptor proteins. SARM1 is a critical protein involved in axonal degeneration, particularly in response to injury and neurotoxic insults. It is widely expressed in the brain and neurons. It is also reported to be expressed in photoreceptor cells of the neural retina. SARM1 predominantly localizes in the cytoplasm and, studies show its localization in mitochondria, axons and dendrites. The protein is synthesized with a transit peptide (aa 1-27) and mature protein contains 8 armadillo repeat (ARM) regions, SAM (sterile motif) domains and a TIR (toll-interleukin receptor) domain. The TIR domain facilitates its activation through dimerization. This activation leads to the hydrolysis of NAD+, a process essential for its function as an axon degeneration executor. SARM1 has been implicated in inflammatory responses particularly in the central nervous system. It can influence the activation of microglia, the immune cells of the brain, and is involved in the regulation of neuroinflammatory processes. This suggests its potential role in diseases characterized by chronic inflammation, such as multiple sclerosis and Alzheimer′s disease. This mouse monoclonal 10G2 antibody has been validated and cited for Western blotting, Immunohistochemistry, Immunoflourescence and Immnuoprecipitation. Studies conducted with this antibody suggests that SARM1 also plays multiple roles in neuronal morphogenesis and, it is specifically required for proper initiation and elongation of dendrites, axonal outgrowth, and neuronal polarization. These functions likely involve Sarm1-mediated regulation of microtubule stability, as Sarm1 influenced tubulin acetylation. (Ref.: Chen, C-Y., et al. (2011).
