General description
Apelin receptor (UniProt P35414; also known as Angiotensin receptor-like 1, G-protein coupled receptor APJ, G-protein coupled receptor HG11) is encoded by the APLNR (also known as AGTRL1, APJ) gene (Gene ID 187) in human. Apelin is an angiotensin converting enzyme 2 (ACE2) substrate that functions as an inotropic and cardiovascular protective peptide. Apelin exerts its physiological effects via its receptor APJ, a seven-transmembrane G protein-coupled receptor that shares significant homology with the Ang II type 1 receptor (AT1R). Apelin/APJ signaling plays an important role in a wide range of pathophysiological effects in the cardiovascular system. Apelin levels are reduced in patients with essential hypertension independent of left ventricular systolic and diastolic dysfunction. Treatment with Apelin has been shown to lower systolic blood pressure levels in hypertensive rats via an NO-dependent mechanism. APJ is a 7-transmembrane (a.a. 27-51, 67-91, 101-125, 145-166, 201-221, 245-271, 285-308) GPCR, having a short N-terminal extracellular portion (a.a. 1-26) and a long C-terminal cytomplasmic tail (a.a. 309-380).

G protein-coupled receptors (GPCRs) activate heterotrimeric G proteins by mediating a GDP to GTP exchange in the Gα subunit. This leads to dissociation of the heterotrimer into Gα-GTP and Gβγ dimer. The Gα-GTP and Gβγ dimer each regulate a variety
