General description

Myeloperoxidase (EC 1.11.2.2; UniProt P05164) is encoded by the MPO gene (Gene ID 4353) in human. Myeloperoxidase/MPO is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a 745 a.a. single chain precursor with a signal peptide sequence (a.a. 1-48), myeloperoxidase is subsequently cleaved into a light chain (a.a.165-278) and a heavy chain (a.a. 279-745). The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. MPO catalyzes hydrogen peroxide-dependent oxidations and the formation of hypochlorous acid that is central to the microbicidal activity of neutrophils.

Biochem/physiol Actions

This monoclonal antibody reacts with majority cell types of the myeloid lineage, including myeloblasts, neutrophils, and monocytes. It does not react with human lymphocytes, thrombocytes, or erythrocytes. 96% of all acute myelogenous leukemias (AMLs) are found positive, 100% of all acute lymphoblastic leukemias (ALLs) are found negative when stained with this monoclonal antibody.

Purified by Ion-Exchange Chromatography
Purified mouse conjugate IgG2aκ monoclonal antibody in buffer containing 20mM Tris, 150mM NaCl, pH8.0 with 1% BSA (w/v), and 0.1% sodium azide.

Preparation Note

Stable for 1 year at 2-8°C from date of receipt.

Analysis Note

Evaluated by Flow Cytometry in human PBMCs.

Flow Cytometry Analysis: 0.01 mL of this antibody detected Myeloperoxidase in human peripheral blood monocytes (PBMCs).
