General description
Glutamyl aminopeptidase (UniProt: P16406; also known as EC:3.4.11.7, CD249, Aminopeptidase A (AP-A), BP-1/6C3 antigen, EAP, gp160) is encoded by the Enpep gene (Gene ID: 13809) in murine species. Aminopeptidase A/ENPEP is a type II integral membrane endopeptidase characterized by a large extracellular C-terminal domain that contains a zinc-binding motif typical of zinc-dependent metallohydrolases. Aminopeptidase A/ENPEP is involved in the regulation of blood pressure and blood vessel formation by cleaving N-terminal glutamate residues from peptides such as angiotensin II, thereby modulating the renin-angiotensin system. It also plays a role in the final digestion of peptides generated from the hydrolysis of proteins by gastric and pancreatic proteases, and is involved in processing various peptide hormones, neuropeptides, and chemokines. Aminopeptidase A/ENPEP is expressed at high levels in the epithelial cells of the kidney glomerulus and proximal tubule cells. Its dysregulation has been implicated in hypertension and cardiovascular diseases due to its role in the renin-angiotensin system. Recent studies have also suggested its potential involvement in cancer progression and immune modulation, highlighting its expression as a predictor for immune checkpoint inhibitor therapy.
