General description
Argonaute(Ago)蛋白是一个约95 kDa的蛋白家族,可直接与成熟的miRNA结合。在哺乳动物中,有四种Ago蛋白(Ago1-4)。该蛋白家族是大多数哺乳动物miRNP的重要组成部分,它们在小RNA指导的调控途径中具有重要功能。Ago家族由两个亚科组成,Ago和Piwi,其包含两个称为PAZ和PIWI的特征结构域。这两个结构域可与成熟miRNA的不同部位发生相互作用。PAZ域可与miRNA的3’端作用,PIWI结构域可与miRNA的5’磷酸盐和5’近端区域作用,以指导靶标mRNA的识别。
