General description

RPA (RP-A p32) 是一种异源三聚体蛋白复合物,可特异性结合单链 DNA (ssDNA)。它由三个亚基组成:RPA1 (70 kDa)、RPA2 (32 kDa) 和 RPA3 (14 kDa)。RPA 在 DNA 复制中发挥多种作用。DNA 复制开始时,RPA 被加载到染色质上,随后加载 DNA 聚合酶和其他复制蛋白来启动 DNA 复制。复制开始后,RPA 随复制叉移动,稳定 ssDNA 并协助 DNA 合成。除了其复制功能外,RPA 还在损伤修复和重组中发挥重要作用。当细胞进入 S 期时,32 kDa 亚基被 cdc2 激酶家族磷酸化;以及因响应 DNA 损伤而被 ATM、ATR 和 DNA-PK 蛋白磷酸化。
