一般描述
溴脱氧尿苷（BrdU）是胸苷类似物，当外源添加时，在DNA合成过程中会掺入DNA中。抗溴脱氧尿苷单克隆抗体用于标记已掺入BrdU的增殖细胞。 与使用放射性胸苷掺入来鉴定正在复制的细胞相比，该免疫学检测方案具有多个优势。标记和检测可以在同一天进行，而不是像氚标记的细胞放射自显影所要求的那样等待几天，并且消除了使用多个标本获得最佳曝光时间的必要性。此外，通过流式细胞术分析进行抗溴脱氧尿苷染色可以同时评估多个参数。
特异性
克隆BU-1与溴脱氧尿苷特异性结合，与碘尿苷的交叉反应性为10%（10%）。克隆BU-1不会与氟脱氧尿苷或任何内源性细胞成分（例如胸苷或尿苷）发生交叉反应。

应用
使用经流式细胞术、免疫细胞化学和免疫组织化学验证的小鼠单克隆抗BrdU抗体（克隆BU-1）检测溴脱氧尿苷（BrdU）。
尽管最初报道染色核DNA BrdU掺入乙醇固定细胞中而无酸或碱变性，但该性质后来归因于存在于所用支原体污染的BU-1杂交瘤培养物上清液中的DNase活性（Gonchoroff，N.J.，et al.（1986）.J. Immunol. Methods.93(1):97-101; Greipp, P.R., et al. (1985).Am. J. Hematol.20（3）:289-292）。在使用本产品进行免疫染色之前，请在样品制备过程中加入适当的DNA变性步骤。

自收到之日起在-20°C可稳定保存1年。分装保存以避免反复冻融。为了最大程度地回收产品，需在融化后和取下盖子之前将原始样品管进行离心。
