Smoothelin是平滑肌细胞(SMC)细胞骨架的组成部分。针对smoothelin的抗体是监测SMC分化的有用工具。Smoothelin仅在完全分化(收缩)的SMC中表达。RNA和蛋白质分析揭示了该蛋白的广泛性物种分布。肌成纤维细胞、肌上皮细胞以及骨骼肌和心肌等具有SMC样特征的细胞不含smoothelin。组织切片的共聚焦扫描激光显微镜以及转染研究显示,smoothelin的丝状组织不同于结蛋白或波形蛋白。smoothelin与肌动蛋白应力纤维共定位。在免疫印迹中检测到两种分子量同工型,即对内脏SMC具有特异性的50 kDa同工型和血管SMC中分子量约为100 kDa的同工型。人smoothelin由位于22号染色体上的单拷贝基因编码。MAB3242可用于区分完全分化的收缩SMC于与增殖SMC。smoothelin阳性细胞的分布表明smoothelin表达与平滑肌组织收缩力之间存在相关性。Smoothelin是评估动脉粥样硬化病变的有用工具。

特异性

应用

免疫细胞化学

冷冻和石蜡包埋的组织切片的免疫组化。对于石蜡切片,强烈建议在10 mM柠檬酸盐缓冲液(pH 6.0)中进行高温抗原修复(van der Loop, 1997)。

最佳工作稀释度必须由最终用户进行确定。
