一般描述

整合素β-1（UniProt P05556；也被称为CD29、糖蛋白Iia、GPIIA、整合素VLA-4亚基β、极晚激活蛋白、β多肽）在人体中由ITGB1（也被称为FNRB、MDF2、MSK12、VLA-BETA、VLAB）基因（基因ID 3688）进行编码。整合素β-1是一种130 kDa的跨膜糖蛋白，可与多种整合素α亚基（包括α1、α2、α3、α4、α5和α6）相互作用，形成与特定细胞外基质结合的功能性受体复合物蛋白。整合素受体可调节多种重要的生物学功能，包括胚胎发育、伤口修复、止血和防止程序性细胞死亡。它们还涉及异常的病理状态，例如肿瘤定向的血管生成、肿瘤生长和转移。这些异二聚体受体将细胞质肌动蛋白细胞骨架与细胞外基质和/或相邻细胞中存在的蛋白质进行桥接。整合素与细胞外基质之间的相互作用会导致信号转导途径的激活和基因表达的调节。

应用

免疫沉淀
一个先前批次的产品已从106个细胞当量的裂解液中对VLA（β1）整合素进行了免疫沉淀：2 μg。

流式细胞术：
10 μg/mL先前批次的产品已被用于FC。

在冷冻组织切片中进行免疫组织化学分析：
使用20 μg/mL先前批次产品。由于VLA（β1）的组织分布广泛，通常能够观察到MAB1997与多种细胞类型发生结合。
其不会阻断VLA整合素介导的细胞对基质蛋白的粘附。通过MAB1997激活VLA（β1）整合素信号转导特性还尚未被表征。

蛋白质印迹：
10 µg/mL。对经过SDS变性和加热的样品有效。对还原后的样品无效。

最佳工作稀释度必须由最终用户确定。

抗整合素β1抗体，克隆MB1.2，是一种抗整合素β1抗体，可用于FC、IH、IP和&WB。

质量

已通过蛋白质印迹对C2C12裂解液进行了常规评估。

蛋白质印迹分析：
该批次的产品已以1：1000的稀释度在10 μg C2C12裂解物中检测到整合素β1。
