巨噬细胞和粒细胞表达信号调节蛋白（SIRP）， SIRP是一种跨膜糖蛋白，参与细胞粘附、生长因子或胰岛素诱导的酪氨酸激酶受体信号转导的下调。它们是CD47的免疫球蛋白样细胞表面受体。CD47结合对吞噬、肥大细胞活化和树突状细胞活化有负向调控作用，可阻止未成熟树突状细胞成熟，抑制成熟树突状细胞产生细胞因子。

大鼠腹腔巨噬细胞。

应用

免疫组化：
冷冻切片和福尔马林固定，石蜡包埋组织切片（需要胰蛋白酶或蛋白酶预处理）。

蛋白质印迹：
该抗体的一个先前批次被用于蛋白质印迹。

流式细胞术：
使用先前批次的1:100-1:200稀释液。（使用10 μL工作稀释液，细胞数量10^6，体积100 μL）。

最佳工作稀释度必须由最终用户进行确定。

生化/生理作用

MAB1407P识别髓细胞和神经元选择性表达的大鼠SIRP（信号调节蛋白）。该单克隆结合在SIRP上与ED9不同的表位上。它不阻断CD172a和CD47的相互作用。
标记大多数巨噬细胞，包括常驻腹膜、肺泡和活化的巨噬细胞。对其他巨噬细胞的识别仅限于偶尔的Kupffer细胞。此外它能标记粒细胞和树突状细胞群。它还广泛地标记大脑。
预期与大鼠和小鼠发生交叉反应。

外形

免疫亲和纯化
纯化的小鼠单克隆IgG2a，溶于含0.09%叠氮化钠的缓冲液中。

制备说明

自收到之日起以未稀释的等分试样形式，在2-8°C可稳定保存6个月。

分析说明

对照
巨噬细胞、单核细胞、淋巴器官。
通过免疫细胞化学对巨噬细胞/粒细胞进行常规评估。
