1的建议ELISA方案。在0.1 M碳酸盐缓冲液pH值9.6中用浓度为5μ g/ml的单克隆抗IL-8抗体（Chemicon目录号MAB1044）包被高容量微量滴定孔（例如Nunc maxisorp），在4ºC下过夜。
2.用PBS+0.05%吐温20（PBST）清洗孔3次。
3. 向孔中添加样品。在湿盒中室温培养1小时。用PBST清洗3次。
4.在PBST中按1：500稀释兔抗IL-8（Chemicon目录号AB1840）并添加100 μL 至孔中。在室温下培养30分钟。用PBST清洗3次。
5. 在PBST中将抗兔IgG-HRP稀释至适当水平并添加100 μL至孔中。在室温下孵育30分钟。用PBST清洗3次。
6.将100μ L TMB色原添加至每个孔中。培养5-10分钟，并用50 μL 2M硫酸终止反应。
注：
抗兔IgG HRP必须吸附鼠免疫球蛋白，应检查与包被板是否无反应性。
可以通过向稀释剂中添加蛋白（例如脱脂奶粉）来减少非特异性结合。
可通过调整培养时间和试剂浓度来改变检测灵敏度。
