General description
The protein named ORM1-like protein 3 (ORMDL3), and encoded by the human gene ORMDL3 belongs to a novel gene family of endoplasmic reticulum membrane proteins that are highly conserved and ubiquitously expressed in adult and fetal tissues. ORMDL3 is a negative regulator of sphingolipid biosynthesis and may indirectly regulate endoplasmic reticulum-mediated calcium signaling. Also in lung tissue, ORMDL3 is an inducible epithelial protein that regulates metalloproteases, chemokines, oligoadenylate synthetases and activates activating transcription factor 6 (ATF6), a critical protein in the unfolded protein response immune response pathway which plays an important role in our response to infections and pathology. The discovery of the link between ORMDL3 and the unfolded protein response pathway may be part of the mechanism of how ORMDL3 is involved in the asthmatic response and regulation. Defects and polymorphims in ORMDL3 gene are linked with Asthma and with asthmatic response profiles.
