一般描述
Tristetraprolin（TTP，Nup 475，TIS11，G0S24或ZFP36）是CCCH串联锌指蛋白家族的成员。是一种作为反式作用因子的核RNA结合蛋白。它参与细胞因子基因的转录后调节，最主要的是肿瘤坏死因子α（TNF-α）。在被脂多糖激活后，TTP在3′-非翻译区（3′-UTR）的富含AU的结构域结合TNF-αmRNA并诱导腺苷酸化，从而使mRNA不稳定并标记其降解。为了产生这种效应，TTP将其他反式作用因子募集到其C末端结构域，并将mRNA衰变酶募集到其N末端结构域。TTP通过促分裂原活化蛋白激酶激活的蛋白激酶2（MK2）和p53驱动的磷酸化而失活。因此，TTP/MK2调节TNF-α的生物合成并介导涉及TNF-α的炎症反应。在小鼠中敲除TTP会产生关节炎，脱发，皮炎，以及与TNF-α过表达相关的一系列其他表型。
